Taxi around an hour, probably; or a taxi to the Ashdod central bus station and then a bus, total around two hours.
There is no sherut from Ashdod to Jerusalem. A taxi will cost 300 shekels one way (possibly with a surcharge from the port, I don't know). Not my estimate; official Transport Ministry fare! And there is a small surcharge - as with all taxi fares anywhere in the country - for more than two passengers.
There is a shuttle from the port to Ashdod center where you can take a bus. The limitation is the hours the shuttle runs, to return to the port. It takes about 1.5 hours to reach Jerusalem by taxi, but you would need over 2 hours should you decide to go by bus. Return to the port only by shuttle as only one Ashdod taxi company is authorized to enter the port.some shipping lines lines offer transportation only to/from Jerusalem which may be worth your while for budgetary /time constraints. Check with the cruise if this is an option.
If you're arriving Friday afternoon or Saturday your only option is taking a taxi. Otherwise you may have public transport, but it'll take more than two hours. Sounds like waste of time. You may want to consider renting a car for the day as well. A good option for about $30 if you don't mind driving.
The OP is definitely not arriving on Saturday ( they are in Haifa that day) BUT it could be on a Friday! A car rental is NOT a good idea. You can waste 1 hour picking up and than another hour returning the rental. Furthermore, the rental will not be next to the port, so they'll first have to get to the agency! ( and then from the agency back to the port) ( and if it is a Friday, agencies are closed by mid day)
