We are in the process of booking our Puglia trip -- We are basing near Pezze di Greco which seems to be a good base for these areas.

I would omit Lecce--look at google maps or the viamichelin website to determine drive times and how much time you want to spend in a car.

Do you want to stay in a town or rural setting?

I would stay in the Itria valley or in Matera, or split it 2-1.

As LuvGoin says, Lecce is the outlier in this - you can’t do everything. Go to the Itria Valley: Locorotondo, Alberobello, or even Ostuni for two nights (if you want the sea then perhaps Polignano a Mare or a masseria near Fasano), and have a night in Matera as you drive north.
